SS-31 vs MOTS-c: Mechanisms and Evidence Compared

Dissolv catalog vial labeled SS-31 10MG and Research Use Only

SS-31 and MOTS-c are different molecules with different research histories. SS-31, also called elamipretide, is a synthetic four-amino-acid peptide studied for interactions with mitochondrial membranes. MOTS-c is a 16-amino-acid peptide encoded within mitochondrial DNA and investigated in metabolic signaling. Their shared connection to mitochondria does not establish equivalent effects or a proven reason to combine them.

This comparison explains what researchers measured, how the human evidence differs, and why a research-material listing must be kept separate from an approved medicine. It provides no dosing or personal-use guidance.

SS-31 vs MOTS-c: the key differences

  • Identity: SS-31 is a synthetic tetrapeptide. MOTS-c is a mitochondrial-derived peptide with 16 amino acids.
  • Mechanisms investigated: SS-31 research includes cardiolipin and inner-membrane organization. MOTS-c research includes AMPK-linked metabolism and communication between mitochondria and the nucleus.
  • Human evidence: Elamipretide has undergone controlled clinical trials. Important MOTS-c studies measured the body's own peptide in humans while testing administered peptide in mice.
  • Regulatory distinction: The FDA approved the specific elamipretide medicine Forzinity for a defined Barth syndrome indication in 2025. That approval does not cover Dissolv's SS-31 research material.

What does SS-31 research investigate?

Mitochondria contain an extensively folded inner membrane. Its folds, called cristae, help organize the machinery involved in energy production. Cardiolipin is a lipid associated with that membrane.

In a foundational 2013 study, Birk and colleagues investigated SS-31 binding to cardiolipin and examined mitochondrial changes in experimental kidney ischemia and reperfusion, where blood supply is interrupted and restored. The work connected SS-31 with preservation of mitochondrial structure and recovery of ATP, a molecule cells use to transfer energy. These were biochemical and preclinical findings under specified experimental conditions. Read the Birk et al. study.

That provides a reason to study SS-31 in mitochondrial biology. Describing it as universally “repairing mitochondria” goes beyond what a particular model can establish. The measured tissue, type of injury, comparator and observation period all matter.

What does MOTS-c research investigate?

The 2015 discovery paper identified MOTS-c within the mitochondrial 12S ribosomal RNA region. Cell experiments linked it with changes in folate-related metabolism, accumulation of the metabolic intermediate AICAR, and AMPK activation. AMPK is a cellular energy-sensing pathway. Mouse experiments examined metabolic outcomes, including insulin resistance associated with diet and age. Read Lee et al., 2015.

A separate 2018 study reported that metabolic stress prompted MOTS-c to move into the nucleus in an AMPK-dependent manner and influence gene expression. This makes mitochondrial-to-nuclear communication a distinct research question, rather than simply another description of cardiolipin binding. Read Kim et al., 2018.

These findings describe pathways and experimental observations. They do not establish a predictable increase in a person's energy, weight loss or longevity from externally supplied MOTS-c.

Compare the research materials

  • SS-31 10mg: a sealed vial of lyophilized research material, with a listed quantity of 10 mg per vial. The studies above focus on cardiolipin and mitochondrial-membrane questions.
  • MOTS-c: a separate sealed, lyophilized research-material vial. Its research history concerns mitochondrial-derived signaling; confirm the current listed fill quantity before ordering.

For qualified laboratory projects, compare the exact molecular identity and request the relevant lot documentation before choosing material. A shared research area does not make these compounds interchangeable.

Why human MOTS-c measurements are not treatment trials

A frequently cited 2021 paper combined several kinds of evidence. Its human component measured endogenous MOTS-c, meaning peptide already produced by the body, around an exercise session. Its administered-peptide experiments involved mice and assessed physical performance and other laboratory outcomes. Read Reynolds et al., 2021.

The distinction is essential. A change in a person's naturally occurring peptide after exercise does not show what happens when that person receives a manufactured peptide. Calling the entire paper a human MOTS-c treatment study would misrepresent its design.

FDA's briefing document for its July 2026 compounding advisory meeting reported that its literature search had not identified clinical studies evaluating administration of MOTS-c-related substances in humans. It also identified unresolved safety questions. This statement concerns the evidence reviewed by FDA and should not be converted into a claim that all possible research has been exhaustively excluded. Read FDA's MOTS-c evaluation.

Elamipretide has clinical evidence, with important limits

On September 19, 2025, FDA granted accelerated approval to Forzinity, an elamipretide injection, to improve muscle strength in adults and children with Barth syndrome weighing at least 30 kilograms. The approval used improvement in knee-extensor muscle strength, an intermediate clinical endpoint; continued approval may depend on confirmatory evidence. See the approved prescribing information.

Results in other conditions must be read separately. The 218-participant MMPOWER-3 trial in primary mitochondrial myopathy did not meet its primary endpoints for walking distance and fatigue at 24 weeks. That result prevents treating the broader clinical literature as uniformly positive. Read the MMPOWER-3 randomized trial.

Neither the Barth syndrome approval nor a laboratory mechanism establishes an anti-aging benefit in healthy people. Dissolv's research SS-31 listing is not the approved Forzinity formulation and should not be represented as an interchangeable treatment.

Does the evidence support combining SS-31 and MOTS-c?

The studies discussed here do not establish a head-to-head winner or a clinically validated combination. Different mechanisms can generate a research hypothesis, but a hypothesis still needs direct testing. Claims that one peptide should be used first, that they reliably work together, or that a “stack” is superior require evidence specific to those questions.

Catalog references for qualified researchers

Dissolv maintains separate listings for SS-31 10mg research material and MOTS-c research material. Use each listing's identity and lot-specific documentation when evaluating material for an authorized laboratory project. Product photographs and nominal vial quantities do not establish identity, purity or clinical equivalence.

Bottom line: SS-31 and MOTS-c belong to different lines of mitochondrial research. A useful comparison keeps molecular identity, experimental model and clinical evidence separate. Dissolv research materials are for laboratory research only and are not intended for human or veterinary use.
