NAD+ turns up in every longevity conversation, usually surrounded by claims the science doesn't quite support. Here's what it actually is.

Not a peptide

Worth stating first: NAD+ is not a peptide. It's a coenzyme — a helper molecule your cells use to run chemical reactions. It sits in the same product category by association, not by chemistry.

What it does

NAD+ stands for nicotinamide adenine dinucleotide. Its central role is in energy production: it shuttles electrons during the process that converts food into usable cellular energy.

Think of it as a rechargeable carrier. It picks up electrons, drops them off, and cycles back. Without enough of it, that production line slows down.

Why longevity researchers care

Two observations drive the interest:

  1. NAD+ levels decline with age. This is well documented across tissues.
  2. It's required by sirtuins — a family of proteins involved in DNA repair and cellular maintenance that have been studied extensively in aging research. Sirtuins consume NAD+ to function.

So the hypothesis is straightforward: if NAD+ falls and repair machinery needs it, does restoring levels help?

Where the honest uncertainty sits

That hypothesis is genuinely interesting and genuinely unproven in humans at scale. The gap between "levels decline with age" and "raising levels extends healthy lifespan in people" is enormous, and it has not been closed.

Much of the encouraging data is from cell culture and animal models. Human trials exist but are generally small and short.

The precursor question

You'll also see NMN and NR discussed. These are precursors — molecules the body converts into NAD+. Whether direct administration or precursor supplementation is more effective is an open research question, not a settled one, regardless of how confidently it's marketed.
